Özet
Tanacetum nitens (T. nitens) is an endemic plant species occurring in Türkiye and has attracted interest as a potential source of bioactive secondary metabolites. However, its phytochemical composition and biological activities have not been sufficiently investigated. Therefore, this study aimed to evaluate the extraction yields, total phenolic and flavonoid contents,
and antioxidant capacities of dichloromethane (DCM) and ethyl acetate (EtOAc) extracts of T. nitens. Materials and Methods: The aerial parts of T. nitens were extracted with DCM and EtOAc using ultrasound-assisted extraction. The extraction yields and total phenolic and flavonoid contents of the extracts were determined using the Folin–Ciocalteu and aluminum chloride colorimetric methods, respectively. The antioxidant capacities of the extracts were evaluated using the ferric reducing antioxidant power (FRAP) and 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical-scavenging assays. All analyses were performed in triplicate, and the results were expressed as mean ± standard deviation.
Results: The extraction yields were calculated as 12.83% for the DCM extract and 11.89% for the EtOAc extract. The total phenolic content of the EtOAc extract (32.25 mg GAE/g) was higher than that of the DCM extract (30.03 mg GAE/g), whereas the total flavonoid content of the DCM extract (80.35 mg QE/g) was higher than that of the EtOAc extract (73.32 mg QE/g). According to the FRAP assay, the antioxidant capacity of the EtOAc extract (23.57 mg TE/g) was higher than that of the DCM extract (18.50 mg TE/g). Based on the DPPH the EtOAc extract exhibited a stronger antioxidant profile, with a lower IC50 value (0.2826 mg/mL).

ABSTRACT
Introduction and Purpose:
Discussion and Conclusion: T. nitens extracts demonstrated significant antioxidant potential.
Although the DCM extract had a higher total flavonoid content, the EtOAc extract exhibited
superior antioxidant activity in both tests. These findings support further investigation of T.
nitens as a promising source of antioxidant compounds and bioactive secondary metabolites.
