Özet

Prostate cancer remains one of the leading induces of cancer-related mortality among men worldwide, necessitating the development of safer and more effective therapeutic strategies. Naturally derived bioactive compounds have recently aquired attention for their potential anticancer properties with reduced systemic toxicity. Rosmarinic acid (RA), a phenolic ester abundant in Rosmarinus officinalis and Mentha spicata, shows diverse pharmacological activities, including anti-inflammatory, antioxidant and anticancer effects. This study examined the cytotoxic and antioxidant effects of RA, cisplatin, and their combination (RA + Cis) on PC-3 and LNCaP human prostate cancer cell lines. Cytotoxicity was evaluated using the MTT assay, while total antioxidant capacity (T-AOC) was measured spectrophotometrically. Additionally, caspase-3 expression was examined via immunofluorescence staining to assess apoptotic responses. RA demonstrated a mild proliferative effect at 48 h, whereas cisplatin exhibited significant cytotoxicity in both cell lines. Combined treatment (RA + Cis) slightly reduced cell viability compared with cisplatin alone, although not statistically significant (p > 0.05). T-AOC levels increased with higher concentrations of both agents, with the RA + Cis combination showing the highest antioxidant response (0.426 ± 0.003 mM/Total Prt, p < 0.05). Immunofluorescence analysis revealed elevated caspase-3 expression in all treated groups, indicating activation of apoptotic pathways.
