Özet

Accurate and rapid detection of parathyroid hormone (PTH) is essential for the diagnosis and management of calcium-related metabolic disorders; however, conventional analytical methods often require complex instrumentation and time-consuming procedures. To address these limitations, we report a graphene-based fluorescence aptasensor that enables simple and efficient detection of PTH via a quenching–recovery strategy. The system integrates a FAM-labeled DNA aptamer with reduced graphene oxide (RGO), exploiting the strong affinity of nucleic acids toward graphitic surfaces and the superior quenching capability of RGO. In the absence of the target, the FAM-labeled DNA aptamer is adsorbed onto the RGO surface, resulting in significant fluorescence suppression. Upon introduction of PTH, specific binding induces a structural rearrangement of the aptamer, weakening its interaction with RGO and restoring fluorescence. This target-responsive behavior forms the basis of a sensitive detection mechanism. The analytical performance of the platform was systematically evaluated under optimized conditions, revealing a wide dynamic response toward PTH with a linear range of 1–200 ng/mL. The calculated LOD and LOQ were 3.97 and 12.03 ng/mL, respectively. The sensing system maintained reliable signal discrimination in the presence of structurally diverse interfering species and demonstrated effective performance in human serum samples after appropriate dilution, indicating its compatibility with complex biological matrices. By combining a straightforward design with target-induced signal modulation, this study presents a practical and scalable sensing strategy.
